| dc.description | The scrapie disease of sheep is a fatal neurodegenerative disease of the central nervous system. This disease is known for almost 300 Years. However, in Cyprus it has first diagnosed in sheep in 1985. The infectious agent of the disease consists of a modified isoform of the normal cellular protein PrPc. Polymorphisms in the PRNP gene of sheep, located at codons 136,154,171, have been associated with the development of resistance or susceptibility to scrapie disease. The present literature review, is divided into five main chapters. The study begins by explaining relevant concepts, such as definitions, historical data and georgraphical spread of the scrapie disease, such as modes of transmissions, symptomatology, diagnostic methods, as well as the public health risk of the disease. In the next chapter, the nature of the causative agent responsible for the disease was identified as well as the mechanism of conversion of the normal protein to the infectious protein. In the fourth chapter, the genotypic composition of the PRNP gene at codons 136, 154, 171, the polymorphisms of the gene and the investigation of the effects of the gene on sheep preformation were studied. Finally, chapter five describes scientific information on legislation and genetic improvement programs for the eradication and development of resistance to scrapie disease. | en_US |
| dc.description.abstract | Η τρομώδης νόσος των αιγοπροβάτων είναι μια θανατηφόρος, νευροεκφυλιστική νόσος του κεντρικού νευρικού συστήματος. Η νόσος αυτή είναι γνωστή εδώ και σχεδόν 300 χρόνια ενώ στην Κύπρο διαγνώστηκε για πρώτη φορά στα πρόβατα το 1985. Ο μολυσματικός παράγοντας της ασθένειας αποτελείται από μια τροποποιημένη ισόμορφη της φυσιολογικής κυτταρικής πρωτεΐνης PrPC. Οι πολυμορφισμοί στο γονίδιο PRNP των προβάτων που βρίσκονται στα κωδικόνια 136,154,171 έχουν συσχετιστεί με την ανάπτυξη ανθεκτικότητας ή ευαισθησίας στην τρομώδη νόσο. Η παρούσα βιβλιογραφική ανασκόπηση διαιρείται σε πέντε βασικά κεφάλαια. Η μελέτη αρχίζει με την επεξήγηση σχετικών εννοιών, όπως σαφείς ορισμούς, ιστορικά στοιχεία και γεωγραφική εξάπλωση της Τρομώδους νόσου. Στο δεύτερο κεφάλαιο αναλύονται γενικά στοιχεία της νόσου, όπως τρόποι μετάδοσης, συμπτωματολογία, διαγνωστικές μέθοδοι καθώς και η επικινδυνότητα της νόσου για τη δημόσια υγεία. Στο επόμενο κεφάλαιο καθορίστηκε η φύση του αιτιολογικού παράγοντα που ευθύνεται για τη νόσο, καθώς και ο μηχανισμός μετατροπής της φυσιολογικής πρωτεΐνης σε μολυσματική. Στο τέταρτο κεφάλαιο μελετήθηκε η γονοτυπική σύνθεση του γονιδίου PRNP στα κωδικόνια 136,154,171 , οι πολυμορφισμοί του γονιδίου και η διερεύνηση των επιδράσεων του γονιδίου στις αποδόσεις των προβάτων. Τέλος, στο πέμπτο κεφάλαιο περιγράφονται επιστημονικές πληροφορίες σχετικά με τις νομοθεσίες και τα προγράμματα γενετικής βελτίωσης για την εκρίζωση και ανάπτυξη ανθεκτικότητας για την τρομώδη νόσο. | en_US |
