Abstract: Ascorbate oxidase (AO) is an apoplastic enzyme in plants that uses oxygen to catalyze the oxidation of ascorbate (ASC) to the unstable radical monodehydroascorbate (MDHA) which rapidly disproportionates to yield dehydroascorbate (DHA) and ASC, and thus contributes to the regulation of the ASC redox state. Here, we report that in vivo lowering of the ASC redox state by increasing AO expression in transgenic tobacco (Nicotiana tabacum L. cv. Xanthi) plants has no distinct effect on the expression levels of genes involved in ASC recycling under normal growth conditions. However, transgenic plants show increased sensitivity when treated with various abiotic stress agents producing reactive oxygen species (ROS), as well as to fungal infection with Botrytis cinerea. RNA blot analyses suggest that this response correlates with a general suppression of the plant’s detoxifying capacity as demonstrated by lower expression levels of ASC recycling genes. Our current findings, combined with previous studies which showed the contribution of AO in regulating the ASC redox state (Planta 216: 918-923, 2003), suggest that decrease of the ASC redox state has an indirect effect on the plant’s capacity to withstand oxidative stress imposed by abiotic and biotic stress agents.
